DMAA was patented by Eli Lilly in 1944 as a nasal decongestant called Forthane. It was produced when many decongestants such as dl-amphetamine or racemic amphetamine were abused for their psychoactive properties. It is a small molecule that shares structural similarity to amphetamines.
Cerebral hemorrhage is a well-known complication of amphetamine use. Two cases of cerebral hemorrhage after DMAA use were reported in 2010.
The 3 cases in this article were also associated with DMAA used as a recreational stimulant. None of the patients had any history or signs of head trauma. The stated doses of DMAA in party pills sold in New Zealand range between 50 and 300 mg per pill. The products are often combined with caffeine.
A recent small volunteer study showed that 75 mg of DMAA ingested orally produces a significant increase in systolic blood pressure 30 to 90 minutes after oral administration. The effect appears dose related. Combination with caffeine produced an additive increase in systolic blood pressure. Cerebral hemorrhage may be a result of a spike in blood pressure, though in patient 2 there was evidence of possible drug-related vasculitis.
Three manufacturer-funded human studies of DMAA-containing supplements have recently been published in the nutrition science literature. They all originate from 1 group of researchers and were published in 1 open-access journal. They studied the cardiovascular and metabolic effects of named dietary supplements in 3 small placebo-controlled trials. These trials have been put forward as evidence of safety for human consumption. None described the actual dose of DMAA administered to trial subjects, so it is difficult to draw any conclusions. Mild increases of pulse rate and blood pressure were observed in the studies.
Safety concerns about this compound have escalated after recent reports of DMAA-induced cardiomyopathy and the deaths of 2 servicemen linked to dietary supplements containing DMAA.
Methylhexaneamine has not been studied intensively and its pharmacological profile has not been evaluated since Eli Lilly filed its patent in 1944
A 2012 Dutch literature study on the regulatory status of methylhexamine concluded that food supplements with > 4 mg methylhexamine are pharmacologically effective and therefore require licensing as a medicine. The authors deduce that oral methylhexamine acts as a bronchodilator (above 4 mg), increases heart rate (above 50–75 mg), and increases blood pressure (above 100 mg). Serious adverse effects are expected for oral dosages above 200 mg.
Anecdotal reports indicate extremely high doses of methylhexanamine combined with caffeine and alcohol may be harmful.

Dat kun je niet stellen want er is nauwelijks wetenschappelijk onderzoek gedaan naar de werking van DMAA in mensen. In contrast met amphetamine onderzoek is het een wereld van verschil.
